Researchers uncover new pathways in bacterial intercellular competition

Apr. 8, 2013 ? There's an epic battle taking place that's not on the national radar: intercellular competition. While it's not an Olympic event, new research from UC Santa Barbara demonstrates that this microscopic rivalry can be just as fierce as humans going for the gold.

Christopher Hayes, UCSB associate professor of molecular, cellular and developmental biology, along with postdoctoral fellow Sanna Koskiniemi, graduate student James Lamoureux, and others, examined the role certain proteins, called rearrangement hotspots (Rhs), play in intercellular competition in bacteria. The findings appear today in the Proceedings of the National Academy of Sciences.

Rhs proteins and related YD-peptide repeat proteins are present in a wide range of bacterial species and other organisms, including human beings, where they help establish communications between neurons in the brain when the visual system is developing. Hayes and his team found that Rhs proteins enable Dickeya dadantii 3937, a phytopathogenic bacterium causing soft rot diseases on many crops, to compete with members of its own kind through touch-dependent killing.

While Rhs have been recognized for more 30 years, their function has been enigmatic. This new research sheds light on the mystery. Rhs proteins possess a central repeat region, characteristically the YD-repeat proteins also found in humans, as well as variable C-terminal sequences, which have toxin activity. C-terminal regions are highly variable between bacterial strains even in the same species, indicating that a wide variety of weapons are deployed.

"Bacteria almost always have a different Rhs toxins," explained Hayes. "No one really knows why, but perhaps the toxins are rapidly evolving, driven by intercellular competition. In essence, these cells are fighting it out with each other. It's like an arms race to see who has the best toxins."

Cellular competition is analogous to that between humans and reflects a scarcity of resources. Like people, bacteria need a place to live and food to eat. "We think these systems are important for bacterial cells to establish a home and defend it against competitors," said Hayes. "In fact, bacteria have many systems for competition. And as we uncover more mechanisms for intercellular competition, we realize this is a fundamental aspect of bacterial biology."

These findings demonstrate that Rhs systems in diverse bacterial species are toxin delivery machines. "We have been able to show that gram-negative (Dickeya dadantii) as well as gram-positive (Bacillus subtilis) bacteria use Rhs proteins to inhibit the growth of neighboring bacteria in a manner that requires cell-to-cell contact," said Koskiniemi, the paper's lead author.

The toxic part of Rhs at the tip (the C-terminal region) is delivered into target cells after cell-to-cell contact. Some toxic tips destroy DNA and others destroy transfer RNA, which is essential for protein synthesis. These toxin activities help the bacteria expressing them to outcompete other members of the same species not carrying an antidote.

This work may help scientists design Rhs-based bacterial probiotics that kill specific pathogens but leave most normal flora unharmed. The research was supported by grants from the National Science Foundation and National Institutes of Health and by fellowships from the Carl Tryggers and Wenner-Gren Foundations.

3 More Cases of New Bird Flu Strain

Three new cases of the new bird flu strain, H7N9, have been reported in China, bringing the total number of cases to 21, according to the World Health Organization. Six of those who were infected have died, but no new deaths have been reported since Friday.

The new patients include a 59-syear-old Shanghai man who is in critical condition, and a 55-year-old Anhui?man who is in stable condition. Another new patient, a 67-year-old Shanghai?man, has a mild case, according to the WHO.

?They?ve already seen some changes that allow it to survive in people,? ABC News chief health and medical editor Dr. Richard Besser told? ?Good Morning America.?? ?The big concern is could this become the next pandemic strain??

More than 530 close contacts of the H7N9 patients? have been monitored, according to the WHO.

The U.S. Centers for Disease Control and Prevention is already working on a vaccine, using the virus?s genetic code? rather than the virus itself ? a first for the agency, according to Nancy Cox, head of the CDC?s influenza division.

Although H7N9 is more easily transmittable from birds to humans than the original bird flu strain, H1N1, Cox said she expects to see limited human-to-human transmission.

Since H7N9 is not as deadly to birds as it is to humans, it will be harder to track because there won?t be large bird kill-offs, Cox said.

?That?s very concerning because you can?t tell where it is without testing the birds directly,? Besser said.

On Friday, a Shanghai market where the virus was detected in pigeons halted live bird sales and slaughtered all poultry, amounting to more than 20,500 chickens, ducks, geese and pigeons, according to China?s? Xinhua News Agency.

?The key to controlling the number of H7N9 patients depends on whether the virus can spread among human beings,? said Wu Fan, director of the Shanghai Municipal Center for Disease Control and? Prevention, at a news conference last Friday, according to Xinhua. ?So far we haven?t found any cases that show this kind of virus can spread from people to people.?

Terrafugia's Transition flying car (or driveable plane) has stayed out of the spotlight since we caught a glimpse of it last year, but AOL Autos recently checked-in with the daring manufacturer to gauge its progress. Sure, the prototype has already scored a VIN from the National Highway Traffic Safety Administration (NHTSA) and an N-number from the Federal Aviation Administration, but it turns out the land /air hybrid is still awaiting complete certification by the NHTSA. Production dates won't be announced until the craft has been rubber-stamped by Uncle Sam, and that isn't expected to occur within the next 12 months. For now, suppliers are being lined up, and it's very likely that another prototype will be fashioned before it hits the assembly line. You might not be able to sit inside your very own Transition soon, but you will be able to see the first model on display at Cape Cod's Heritage Museum this summer.

Telefonica Mulls More Disposals to Pay Down Debt

?Spain's Telefonica is reportedly looking at fresh asset sales to help pay down debt only a few months after it scrapped plans to sell a stake in its Latin American operations.

The company has a target of cutting its EUR51 billion of debt by at least EUR4 billion this year.

Citing people familiar with the situation, Bloomberg News reported that the company's O2 Ireland and Czech Republic subsidiaries are being considered for sale. The company floated a stake in O2 Germany last year, but recently ruled out a similar move for its UK division.

Other options are reported to include Telefonica's 5 percent stake in China Unicom, which has been valued at around US$1.6 billion. That sale could not take place before June though, as the company made a commitment to hold onto its shares for at least a year when it sold a 4.56% stake in China Unicom last year.

The company saw its debt ratings cut last year as ratings agencies worried about the ability of the company to maintain its creditworthiness. Since then a sale of a stake in its German operations and improved performance in Latin America has lifted the financial pressure somewhat.

However, at the moment, no decision has been made and no banks have been hired, the sources added.

North Korea can likely revive reactor in six months, needs years for more bombs

UNITED NATIONS (Reuters) - North Korea can probably restart a mothballed plutonium-producing reactor in six months if it is determined to do so and the site has suffered no major structural damage, but it may take years to produce significant new atom bomb material.

Pyongyang announced on Tuesday that it would revive the aged Yongbyon five-megawatt research reactor that yields bomb-grade plutonium, but stressed it was seeking a deterrent capacity.

Several nuclear experts familiar with North Korea's program said it would probably take the North Koreans about half a year to get the Yongbyon research reactor up and running, provided it has not suffered significant damage from neglect.

The decision to restart the reactor was the latest chapter in an escalating crisis that erupted after Pyongyang was hit with U.N. sanctions for conducting a third nuclear test in February, and the United States and South Korea staged military drills that North Korea viewed as "hostile."

The Yongbyon reactor has been technically out of operation for years. But Siegfried Hecker - a Stanford University nuclear scientist who is believed to have been the last Westerner to visit the Yongbyon nuclear complex - said the Yongbyon research reactor has been on standby since July 2007.

"If they restart the reactor, which I estimate will take them at least six months, they can produce about six kilograms of plutonium (roughly one bomb's worth) per year," Hecker said in an interview published on Tuesday on a Stanford website.

He said that it would take the North approximately three to four years before it could get another 12 kg (26 lbs) of plutonium, which would suffice for two more weapons.

Satellite images published by 38North, a specialist North Korea website, showed new construction activity at the reactor site from early February until the end of March. It said the imagery indicated that construction had begun along a roadway and toward the back of the reactor building.

NO FOREIGN HELP NEEDED, EXPERTS SAY

Isolated North Korea occasionally lets nuclear experts like Hecker into the country, most likely to persuade them that its nuclear capabilities are not imaginary, U.N. diplomats and officials say.

Hecker added that when he last visited North Korea in 2010, he estimated that the country had a stockpile of 24 to 42 kg (53 to 93 lbs) of plutonium, roughly four to eight bombs worth. If the country's February nuclear test used plutonium - which is not clear - the stocks would be about five to six kg lower, he said.

North Korea has repeatedly threatened to use nuclear weapons to attack the United States and its bases in South Korea, but Hecker said he was skeptical about Pyongyang's ability to hit targets on U.S. or South Korean territory.

Olli Heinonen, former head of the U.N. International Atomic Energy Agency's (IAEA) safeguards department, told Reuters he had a similar prediction, though he said it was possible North Korea could have the research reactor running in less than six months.

"We don't know how much preparatory work they've done," said Heinonen, who is currently at Harvard University and has visited North Korea and met with North Korean scientists.

Both Hecker and Heinonen said North Korea could most likely restart the reactor without any foreign assistance, despite U.N., U.S. and other sanctions aimed at curtailing its ability to purchase nuclear and missile technology.

A U.S. official concurred with Hecker and Heinonen.

"North Korea's assertion that it intends to bring Yongbyon back on line can't be easily written off as an insurmountable hurdle," the official said on condition of anonymity.

Mark Fitzpatrick of the International Institute for Strategic Studies think tank in London, however, said there was a possibility that the Yongbyon reactor has been rendered inoperable for unknown reasons.

"It's been a mystery to me why they haven't started it up before this," he said. "The most logical answer is that they couldn't ... But there's no certainty here."

If the reactor is functional, Fitzpatrick said, the half-year timeline for restarting it made sense.

URANIUM ENRICHMENT

IAEA spokeswoman Gil Tudor said North Korea's decision to restart Yongbyon was "another deeply regrettable development, which is in clear violation of U.N. Security Council resolutions."

The Security Council has repeatedly sanctioned North Korea for its nuclear tests and repeated missile launches, and ordered it to abandon both its nuclear and missile programs.

Heinonen said North Korea has already mothballed and restarted the five-megawatt graphite-moderate research reactor before. It shut down the plant after signing the "Agreed Framework," a 1994 deal with the United States under which Pyongyang agreed to freeze Yongbyon in exchange for heating oil and construction of newer light-water reactors.

Pyongyang began to restart the reactor in late 2002 after Washington accused it of secretly developing a parallel uranium enrichment program in violation of the 1994 deal. Washington ceased aid to the North and Pyongyang accused it of reneging on its promise to build the light-water nuclear reactors.

North Korea then expelled all inspectors from the IAEA and in 2003 withdrew from the nuclear Non-Proliferation Treaty. In 2006, it tested its first nuclear device, using plutonium from Yongbyon, followed by two more in 2009 and earlier this year.

Certain technical challenges await the North Koreans. In 2008 they destroyed the Yongbyon reactor's cooling tower as a confidence-building step in U.S.-led multilateral negotiations aimed at reducing tensions on the Korean peninsula.

But the reduction in tensions was short lived. Six-nation aid-for-disarmament talks between the two Koreas, China, Russia, Japan and the United States have been stalled for years.

Heinonen said that either North Korea must build a new cooling tower or create an underground cooling plant, like one that was under construction at a site in Syria that Israel bombed in 2007. Western intelligence sources have said North Korea helped build the Syrian reactor, which the government of President Bashar al-Assad has said was not a nuclear site.

David Albright, a former weapons inspector and head of the Washington-based Institute for Science and International Security think tank, said it was important not to underestimate the nuclear capabilities of the North Koreans or their determination to live up to their word.

"North Korea huffs and puffs a lot, but underneath that they pretty much do as they say," said Albright, who met with North Korean nuclear scientists in Pyongyang in 2011. "They have been saying they want to improve the quality of their nuclear weapons and they may very well do that."

As well as reviving the reactor at Yongbyon, the North's only known source of plutonium for its nuclear arms program, Pyongyang's official KCNA news agency said a uranium enrichment plant would be put back into operation.

Hecker, who visited the enrichment plant in 2010, said North Korea has a good safety record for its five-megawatt research reactor, but he voiced concerns about the new plant it intends to construct.

"I am much more concerned about the safety of the new light-water reactor they are building," he told Reuters without elaborating.
